The present work describes the development and construction of a laser scan microscope using a new phase contrast method. This includes the assembly of the mechanical, optical, and electronic components needed to fundamentally create the microscope. Furthermore, the development of a new imaging method, the matched-masks phase contrast, is presented. The laser beam is focused on an object mounted on a piezo table, with the object being scanned line by line. A photodiode detects the signals reflected from the object. A computer program created with LabView reconstructs an enlarged image of the object from these signals. A video microscope allows for finding the focus at the object level and optimally checking the movement of the scanner. The book is aimed at physicists, engineers, and technical companies for whom microscopy is of importance.
