Is this sound card worthwhile with a DT 99 Pro (250 Ohm)? Do I have to make any settings in the software or on the PC? Do I have to pay attention to which USB port I connect this sound card to?

You mean DT 990 PRO... Yes, that's how I use them and they're great. Fits for music and games, I just have an additional microphone on the headphones. You don't have to set anything special except a switch on the SB itself (L-GAIN-H), just set it to H for high because the 250 ohms have to be driven. USB connection doesn't matter. One problem, however, is the cable supplied at the time (micro USB), which can quickly become loose, apparently known, so I took my own different one.
